What Is Glycine?

Glycine is the simplest amino acid in the human body. Yet this molecular simplicity belies an extraordinary range of biological functions. Glycine is the most abundant amino acid in collagen, a key inhibitory neurotransmitter in the brainstem and spinal cord, a critical substrate in the synthesis of haeme, creatine, and glutathione, and an important modulator of metabolic and sleep function.

A mechanistic study by Kawai et al. confirmed that glycine's sleep-promoting and hypothermic effects are mediated specifically by NMDA receptors in the suprachiasmatic nucleus (the brain's master circadian clock). Ablation of the SCN completely abolished both effects, establishing a precise neurological pathway for glycine's action on sleep architecture.[2]

A 2023 RCT by Thomas et al. found that glycine-rich collagen peptide supplementation (15g, taken 1 hour before bed for 7 nights) in physically active males significantly reduced overnight awakenings measured by polysomnography and improved performance on the Stroop cognitive test the following morning, demonstrating both sleep quality and next-day cognitive function benefits.[3]
